Due to the aggressive nature of cancer, extensive research has been conducted over the past several decades with the goal of developing better therapies that have fewer side effects. One such protein, known as MCL-1 (Myeloid Cell Leukemia-1), has been recently discovered and is a member of the anti-apoptotic BCL-2 family. The overexpression of MCL-1 proteins has been predominantly observed in patients diagnosed with cancer, thereby making it an attractive target for therapeutic intervention. Consequently, substantial research efforts have been directed at inhibiting this protein.

The high expression levels of MCL-1 are recognized as a primary cause for the development of diffuse large B-cell lymphoma, as well as other blood disorders. Moreover, this inhibitor promotes rapid apoptosis and helps to overcome drug resistance, demonstrating a synergistic effect when combined with other treatments. For these reasons, MCL-1 is viewed as a promising candidate in the field of oncology therapy. Although no MCL-1 inhibitors have yet been approved by the FDA for clinical or commercial use, there is an increasing push among scientists and researchers to develop effective inhibitors targeting MCL-1.

Various pharmaceutical companies, along with numerous research centers and universities, are actively pursuing innovative inhibitors designed to combat cancer. These organizations, including Novartis, MD Anderson Cancer Center, Victorian Comprehensive Cancer Centre, Prelude Therapeutics, and Nanfang Hospital at Southern Medical University, are diligently working on understanding the pathogenesis of the MCL-1 protein in order to develop its inhibitors targeting several cancer disorders, including acute myeloid leukemia, non-Hodgkin lymphoma, chronic lymphocytic leukemia, multiple myeloma, and sarcoma.

The market for MCL-1 inhibitors holds immense potential, as these therapies can help overcome challenges associated with conventional treatments currently available in the commercial market, such as those involving chemotherapeutic drugs. At present, both the US and EU dominate the arena of MCL-1 inhibitors, thanks to a growing pipeline of clinical trials.
